As far as I'm aware, Vespa worked for the treasury, and Mathis was an MI6 liason officer ie the man on the ground to act as aid/fixer/back-up etc. I think that Bond suddenly thinks why is she reporting to Mathis and questions this. It's not very clear and has troubled me also.

Bond discovered Le Chiffre's tell(sign that he is bluffing) and Le Chiffre knew this and was able to make Bond think he was bluffing. Bond believes someone has told Le Chiffre and assumes it to be Mathis.

Yeah, except Bond himself told Le Chiffre he'd discoved his 'tell', referring to his eye bleeding beforehand to rattle him.

But that wasn't the tell, was it? Wasn't it tapping his temple?

This part with Mathis threw me the first time I saw it -- Loeff had to explain it to me. Generally, I prefer subtlety in films, but this moment seemed more cryptic than subtle because there really wasn't anything suspicious about Vesper's statement or the idea that Mathis would want to meet her. I almost wonder if some script element might have been cut that would have implicated Mathis just a bit more strongly. Or I just may be getting denser.

In the book, it's clear that the note she supposedly left was 'the crudest possible forgery'.
