My favorite is when Bond is driving and a woman passes him and begins to speed up to chase her - then he realizes that he is on a mission and mutters, "Discipline 007, discipline."

Guess number one goes to: "What, no small talk? No chit-chat?" And Pierce's delivery of that one was absolutely priceless.
Also loved this one from LTK when Dalton introduces Q and Pam Bouvier:
"Miss Kennedy, this is my uncle Q. Q this is my cousin, Miss Kennedy"
Q (taking advantage of the situation to hug and kiss her): "We must be related!" )
The goodness of that was all Desmond's acting. It was too damn funny.
And then the classic:
